ESTIMATION OF PLASMA/SERUM CHLORIDE WITH COULOMETRY
REAGENT
- DILUTING FLUID
COMPOSITION
Glacial acetic acid 100ml
Concentrated nitric acid 6.4ml
Deionized water 1litre
PREPARATION
Mixed 100ml of glacial acetic acid with 200ml of deionized water, add 6.4ml of concentrated nitric acid, and make it up to 1lite with deionized water.
- INDICATOR SOLUTION
COMPOSITION
Unflavored gelatin 6.0g
Thymol blue 0.1g
Thymol 0.1g
Deionized water 1litre
PREPARATION
Dissolved 6.0g of unflavored gelatin, 0.1g thymol blue, and 0.1g hymol in 500ml of dionized water, make up the solution to 1litre with dionized water by gentle heating, when the it fully dissolved, bring the solution to room temperature and make it up 1litre with deionized water. Store at 2-8ºC.
- STANDARD CHLORIDE SOLUTION 100mmol/L
COMPOSITION
Sodium chloride 5.85g
Deionized water 100ml
PREPARATION
Dissolved 5.85g of Sodium chloride in 100ml of dionized water
METHOD
- Measure 0.1ml of serum in test tube
- Add 10ml of reagent 1 to the test tube
- Add 2-3 drop of reagent 2
- Deep in the electrode in the test tube, switch on the machine
- Start the digital display
- There will be a stop of the display reading on machine when the concentration of chloride is indicated
- The same procedure should be repeated with 0.1ml of reagent 3 in other to be sure of the accuracy of the result.
