LOEFFER ALKALINE METHYLINE BLUE

This is a basic dyes used for examination of microorganism structure in the microbiology laboratory, these method is usually appear to smear from cultures. The solution is slower than that of methylene blue in such way lead to slow performance like that of polychromatic staining.

SOLUTION

Methylene blue in 1.5% in 95% alcohol 300ml

Potassium hydroxide             10ml

Distilled water                       990ml

PREPARATION

Measure potassium hydroxide solution into distilled water and mix with methylene blue solution thoroughly filter date and record.

PROCEDURE

Apply the solution for 30 second, wash with water blot carefully and dry with gentle heat.

Examine with microscope.
