On the origin of bacteriology, the culture media used by then are ox Heart or Calf brain. But with the advancement of knowledge of bacteriology, this type of media is drastically disappearing in the microbiology world, and is been replace by water free media or media constituent. Some bacteriology laboratory depends on buying commercially prepared media, while some laboratory prepared the media by themselves. There are many company today in the world that specialized in the preparation of media and sold it to laboratories in the world, like Himedia, Becton Dickinson. Preparation, storage, composition of culture media, sees manufacturer instruction. For better performance of culture media, the manufacturer instruction should be adhere to. When culture media arrive at the laboratory, the potency of the media should be check by the laboratory personnel, by doing that it will give you the idea of qualitative content of the media. There are reason why the potency test of the newly arrived media in the laboratory should be carried on before subject for used in the laboratory, the reason is because the media can develop fault at a place where it was store, or from the manufacturer the media is not properly produce either using less product or using substandard product. In case of examination and the media is found to be dehydrated, distilled or deionized water should be used to rehydrate the water, with ineffective performance will be avoided. Water that is left to stand for a long period of time should not be used because the water must have assimilated gas or chemical from the atmosphere, and if such water is used, it can alter the composition of the media which will result to faulty performance of the media. The effect which can take place is change in the PH of the media. Media should be store in cool and dry place, dry from sun light should be avoided, also care should be taken when handling the media because dampness cause form of solid lump on the media, which even when the media is rehydrated, it will not attained the satisfactory performance.
