These are important anaerobic covering, and it is made up of a gas tight chamber with cemented covering portal and an inlet lock for moving substance in or out the chamber. There are many indicator designs to examine this principle of this organism, the indicator is called anaerobic indicator. The indicator help examine the substance under culture to make sure that anaerobic status is achieved in a desire condition. These task are carried via examination of redox potential (Eh) in the jar, the result which show either it hass been reduce bellow -50mv. When there is a drop in the potential in the jar, then the color of the indicator change to colorless. Another indicator used is biological indicator, this involved the use of obligate micro organism such as pseudomonas aeruginosa and obligate anaerobic micro organism such as clostridium tetani, and they are used to control the internal content mechanism the jar. Pseudomonas is use as a negative control while clostridium is use as a positive control.
INDICATOR
Alkaline methylene blue-glucose solution, the solutions are produce in three forms which are;

0.1M sodium hydroxide 6ml
Distilled water 94ml
- Solution
0.5% methylene blue 3ml
Distilled water 94ml
- Solution
Glucose 6g
Distilled water 100ml
Crystal thylmol little
PROCEDURE
Measure 2ml amount of each of the three solutions in a test tube, boil the solution till colorless develop and place it in an anaerobic jar clamp the lid and proceed as it is described on the anaerobic and cabinet control.
Alkaline glucose broth of pH 8.5 with methylene blue added can be used as an alternative.
