SODIUM/POTASIUM ESTIMATION

REAGENT   A

STOCK SODIUM CHLORIDE STANDARD (1.0M)

COMPOSITION

  1. Sodium chloride 45g
  2. Distilled water 1litre

PREPARATION

Dissolved 58.45g of sodium chloride in 20ml of distilled water, make up the solution to 1litre with distilled water.

ESTIMATION OF PLASMA/SERUM CHLORIDE WITH COULOMETRY

REAGENT B

STOCK POTASSIUM CHLORIDE STANDARD (1.0M)

COMPOSITION

  1. Potassium chloride 55g
  2. Distilled water 1litre

PREPARATION

Dissolved 74.55g of potassium chloride in 20ml of distilled water, make up the solution to 1litre with distilled water.

METHOD

For examination of Na/K, set up 4 tubes this is shown below;

Serum/plasma 0.1ml      
STD 1   0.1ml    
STD 2     0.1ml  
STD 3       0.1ml
  1. make sure that the sample standard preparation is well mixed
  2. The air compressor should be switch on and the air pressure should be adjusted.
  3. The atomizer is channel of introducing deionized water, so make sure that the deionized water is introduce via atomizer.
  4. The gas should be switch on and the flame adjusts when it is adjusted properly, it is expected that it give a sharp fine cone flame.
  5. Filter of both Sodium and Potassium should be place simultaneously in an appropriate place.
  6. Then set the machine to zero using deionized water.
  7. Standard 1 should be place, adjust the potassium and sodium to 2.0 and 120.0
  8. Standard 2 and 3 should check to make sure it displays the same concentration for both potassium and sodium.
  9. Then diluted serum should be introduced and appropriate reading of both potassium and sodium noted and recorded.

NB: Blood sample left standing for more than 4 hours before separating the plasma/serum from the red cell is not good for the test, because there will be a leakage of electrolyte when the sample is left for more than 4 hours.

 

MORMAL RANGE

Potassium             3.5-5.2mmol/L

Sodium                135-146mml/L
